Tissue Culture Glassware
Note: Any contamination (especially by soap) of glassware used in tissue culture can alter the media and ruin the cultures. Glassware must be washed first thing in the morning in order to allow time to cool.
1. Dress Code ( as per HHMI's procedures)
a. Leather shoes or rubber boots are forbidden as do not provide enough protection against hot water spills
b. Buttoned lab coats are mandatory at all times
c. Rubber aprons are to be used when removing hot liquids or decontaminated waste bags.
d. Heavy-duty autoclaving loves (such as NOVATEX non-asbestos high temperature handling mittens from Lab Safety Supply) are to be worn while removing items from dishwasher.
e. Rubber or neoprene gloves (such as UltraFlex RC-11559 from Lab Safety Supply) are to be worn while handling dirty glassware in order to prevent cuts, contamination, or acid burns.
2. Pre-loading into dishwasher
a. Remove all stickers by either pealing or by using a razor blade.
b. Remove any glue residues or ink with acetone.
c. Check dishwasher and DO NOT RUN DISHWASHER WITH SOAP.
3. Load the tissue culture glassware into the dishwasher (see dishwasher) and run without soap.
4. After cycle is finished, hand rinse the bottles five or sink times under deionezed water five or six times.
a.
T.C. glassware has t be extremely clean.
b. Do not leave glassware in sink. Soap might get into it.
5. Soak the caps in water to remove any left-over media
6. Sterilize the bottles (see Autoclave)
a. Set the timer for 40 minutes sterilizing and 30 minutes drying.
7. After both the bottles and caps are washed, tighten the caps onto the bottles all the way to prevent de-sterilization.
